意思是:出了什么问题呢?有什么事情呢?
如果用：
What is wrong with you?
语气过于强烈,表示：你有什么毛病?你有什么问题?
为了更加礼貌和和蔼地回答,用第一句更加合适.
当然,如果你从语法角度分析,两个答句都是可以的.

前有一句带有反问译为“这看起来是问题吗？” 后一句则是疑问句，是对生活现状、状态出现问题而表示的关心。
其实可以的，只不过What seems to be the problem比What is wrong with you?
